Biosynthesis of the peptidoglycan of bacterial cell walls. XI. Formation of the isoglutamine amide group in the cell walls of Staphylococcus aureus.

چکیده
The lipid intermediates in peptidoglycan synthesis in Sfaphylococcus aureus, N-acetyhuuramyl(-pentapeptide)-PP-lipid and N-acetylglucosaminyl-N-acetyhnuramyl(-pentapeptide)-P-P-lipid, act as acceptors of ammonia in an ATP-dependent reaction in which the ar-carboxyl group of glutamic acid is amidated. Procedures have been developed for the separation of the amide and the amide-free forms of the lipid intermediates, and it has been shown that the amide-free lipid intermediate can substitute for the uridine nucleotide substrates in the amidation reaction. Some properties of the reaction are reported.

Bacteriolyses of bacterial cell walls by copper (II) ions and zinc (II) ions based antibacterial results of broth dilution medium method and halo antibacterial test were investigated. From dilution medium method, MIC=625mg/L, MBC=1250mg/L for Cu2+ solution as bactericide action were obtained against Staphylococcus aureus, and also from halo antibacterial test, the high antibacterial effects for...
متن کاملInhibition of cell wall turnover and autolysis by vancomycin in a highly vancomycin-resistant mutant of Staphylococcus aureus.
A highly vancomycin-resistant mutant (MIC = 100 microg/ml) of Staphylococcus aureus, mutant VM, which was isolated in the laboratory by a step-pressure procedure, continued to grow and synthesize peptidoglycan in the presence of vancomycin (50 microg/ml) in the medium, but the antibiotic completely inhibited cell wall turnover and autolysis, resulting in the accumulation of cell wall material a...
متن کاملStaphylococcus aureus peptidoglycan induces histamine release from basophil human leukocytes in vitro.
Whole killed cells, cell walls, and peptidoglycans of Staphylococcus aureus were found to release histamine from human leukocytes and isolated rat mast cells in vitro. The histamine-releasing capability increased in the order of whole bacteria, cell walls, and peptidoglycans. Peptidoglycan was found to release histamine by a nonimmunological mechanism, as demonstrated by release in cells depriv...
متن کاملBiosynthesis of the peptidoglycan of bacterial cell walls. XXI. Isolation of free C55-isoprenoid alcohol and of lipid intermediates in peptidoglycan synthesis from Staphylococcus aureus.
A particulate enzyme system from Micrococcus roseus R2J has been obtained which catalyzes the biosynthesis of a peptidoglycan. The preparation catalyzes the incorporation of L-threonine onto the e-amino group of lysine in the presence of transfer RNA @RNA), supernatant solution, and ATP. Crude l%-L-threonyl-tRNA will substitute for these additional requirements. The threonyl-tRNAs of this organ...
